Hmm… At first that seemed like a re-hash of sneak peek for 10.15 (Sam doing research in secret and Dean almost catching him but Sam playing it off like it’s no big deal.)
Was Dean a little disappointed at first that he won’t get to hang out with Sam? :)
I like that the scene has elements of them being brothers like now Sam has his own room so he doesn’t want his big brother snooping, which is something that Dean totally wants to do. And Dean giving Sam “permission” to stay out all night like he probably did when they were growing up. But something was a bit off.
Jared’s hair getting messy between cuts was distracting. :D
I’d like to think Dean was just playing up the big brother/little brother angle. Another way of saying, “Okay, now I know in advance that you might not be back till morning so I won’t worry if I don’t see you till then. Stay safe.”
However… Does Dean really not suspect anything? Sam’s white lies are not the most convincing. Does Dean have plans of his own so he didn’t want to go to the movies with Sam? On the other hand, he did suggest that they’d stay at the Bunker drinking.
I really doubt something happens between Sam's departure at the end of this sneak peek, and the moment where he meets up with Cass, to make him say that Dean's getting worse. The MoC storyline is a complete mess.
All I can think of is something happens with Dean before this clip. Otherwise you're right and ELI's right as Dean showed no signs that he was affected by the Mark in 10.15/10.16.
I wouldn't count on it. Dabb is one of the better 'Sam writers' and given the right dialog, Sam can be very expository. IF that's case - it may be just as you and BoxManLocke and there will be no explanation to bridge the ending of 10.14 and this episode. I'm looking forward to this episode and I'll think we'll see real progress in both Dean's MoC arc and Cas & Sam's 'covert operation SAVE DEAN' .

In the promo Sam says that Dean is getting worse, but it doesn't seem like it. At least here...

why does Sam need Dean's 'permission' to do anything or was that Dean just being his usual controlling self?

Very sneaky, Sam. Very sneaky. For once, I approve of the lie. Cuz we know dam well that Dean would not let Sam go do that seance.
